DNA marker indicates if ovarian cancer treatment will be successful, study suggests

ScienceDaily (Apr. 3, 2012) ? Researchers and doctors at the North Shore-LIJ Health System and the Feinstein Institute for Medical Research discovered that blood can help determine the best treatment plan for patients with ovarian cancer. More specifically, a genetic marker embedded in deoxyribonucleic acid (DNA), called microRNA, indicates if a patient with ovarian cancer has a benign or cancerous tumor, and that she will benefit from chemotherapy after surgery on the tumor.

This data was presented at the American Association for Cancer Research (AACR) Annual Meeting which was held from?March 31- April 4 in Chicago, IL.

It is estimated that there will be 22,280 new cases and 15,500 deaths this year from ovarian cancer in the United States. Due to lack of adequate screening, the majority of patients with ovarian cancer are diagnosed at stage III (the second-to-last and most devastating stage of cancer), when 70 percent of these patients will die of their disease within 5 years.

"The discovery that microRNAs can help predict the best treatment plan for women with ovarian cancer, who are most likely at stage III of the disease, offers them enormous hope," noted Iuliana Shapira, MD, director of the Cancer Genetics Program at the North Shore-LIJ Health System's Monter Cancer Center. "We can now inform patients at stage III ovarian cancer, if they will have success with chemotherapy following surgery, similar to patients who are at stage 1 disease. This information gives them hope that their disease is curable despite being diagnosed at an 'advanced stage.' It also gives them the strength necessary to undergo chemotherapy, which is a very invasive and toxic therapy necessary to obtain the cure."

Several microRNAs have been found to have links with various types of cancer. The research conducted at the North Shore-LIJ Health System and the Feinstein Institute for Medical Research found that microRNA-195 increased 40 fold during chemotherapy and microRNA-16 increased 80 fold during chemotherapy. These changes may explain why some patients with ovarian cancer have side effects of chemotherapy, why others become cured of cancer as a result of chemotherapy, and why others need ongoing chemotherapy to continue living with the cancer.

"Understanding the changes in microRNA throughout chemotherapy treatment helps us better understand ovarian cancer and how best to treat patients who have this disease," said Annette Lee, PhD, associate investigator at the Feinstein Institute. "The genetic markers we identified allow patients to individualize their own therapy in order to have maximum benefit and minimal side effects. In addition, this knowledge will help researchers develop new treatments for patients with ovarian cancer."

Dr. Shapira adds that, "We applied for a government grant and hope to receive the funds needed to validate these markers allow result in women receiving therapies that are more personalized and match their genetic makeup."

The formal unveiling of LG's Viper this morning may not have piqued your interest, but Sprint loyalists hellbent on maintaining access to an unlimited plan may want to take a second look. Initially pointed out by TechHog, and confirmed to us today by a Sprint spokesperson, the carrier's impending LTE data network will indeed be included on its existing Everything plans. In other words, the Viper -- as well as any other LTE smartphone, Galaxy Nexus included -- will be able to surf the LTE superhighway without limits. To date, the "unlimited" nature of Sprint's data remains a huge differentiator in a world full of hamstrung options, tiers and throttles, and it'll certainly be used to get the attention of heavy users in the months ago. We also reconfirmed that Atlanta, Baltimore, Dallas, Houston, Kansas City and San Antonio are on track to receive LTE in "midyear 2012," with "other markets following in the third and fourth quarters." Huzzah!

'Titanic 3D': The Reviews Are In!

Critics are overwhelmingly impressed with James Cameron's 3-D conversion.

James Cameron has never been known for taking things halfway, and the new 3-D conversion of his epic "Titanic" is no exception.

Cameron and his team spent years adding computer-generated depth to each frame to commemorate the 100th anniversary of the tragedy. When the director screened 20 minutes of footage for journalists last October, they responded with warm reviews, but now the final product is ready to set sail.

Cameron has always stood by 3-D technology for film, through seemingly endless criticism, and now the reviews for his latest 3-D product are coming in as raves.

Drew McWeeny from HitFix doled out high praise for "Titanic 3D," calling it "the very best 3-D conversion of a film that I've seen so far." Though the quality of the 3-D impressed McWeeny overall, he concluded that nothing about the new version of "Titanic" is really that different. "Adding 3-D to that works technically, but I'm not sure it changed anything, one way or another, emotionally," he said.

When converting a film as iconic as "Titanic," expectations for the re-release can be very high. Lou Lumenick from the New York Post applauded Cameron's use of 3-D, saying that it even enhanced the quality of the film. "James Cameron's spectacular new 3-D version of 'Titanic' is everything I'd hoped for, and more," Lumenick said. "He judiciously — and sometimes with great subtlety — uses the technology to make a great film even greater."

Lumenick said he found "Titanic 3D" "less of a strain on the eyes" than Cameron's "Avatar."

While most 3-D movies come with complaints from the critics, post-conversion tends to suffer a worse fate. Scott Mantz from "Access Hollywood," however, wrote that the post work on "Titanic" surpasses even films made for the format. "But what's amazing is that for a movie that was shot in 2-D, 'Titanic' looks amazing in 3-D — almost like it was meant to be in 3-D all along."

Dynamically shot and paced like a thriller, the film has the density and moral prickliness of a good novel.

These people seem so real they might live next door. And they probably do.

Very few movies capture as convincingly as A Separation does the ways in which seemingly honorable decisions can lead to interpersonal conflict -- even disaster.

To say the piercing Iranian film A Separation is about divorce is a bit like saying The Wizard of Oz is about a pair of slippers.

"A Separation" moves beyond one couple's sundering marriage to reveal growing rifts between generations, ideologies, religious mind-sets, genders and classes in contemporary Iran.

"A Separation" is a great movie, a look inside a world so foreign that it might as well be another planet, yet so universal that its observations are painfully familiar to anyone, anywhere.

... subtlety and a sense of profound decency ...

A clever, insightful and heartfelt examination of human frailty that makes watching subtitles seem like a breeze.

It's fast paced, exciting, thrilling, edgy, moving, engaging, and - in its portrait of a justice system almost radically alien to the one I live under - absolutely fascinating.

Although the film might serve as a portrait of Iran's two conflicting social groups as reflected in the Green movement/Ahmadinejad clashes, it is much more about moral contradictions that any society has to face. A masterpiece.

It's the little moments in Farhadi's film that are its most important, speaking every bit as loudly as its big, narrative-driving moments.

If any one film can re-inject life into an entire national cinema, it's A Separation.

You may also find that some aspects of this very foreign story seem disturbingly not all that foreign.

An outsider could see these characters as misguided. That's not the reality however. Farhadi slips us into their shoes and we appreciate each of their perspectives.

Asghar Farhadi has written a superb screenplay and directs it with equal brilliance. He has managed to make a film that says something about the state of affairs in his Iran and weaves that message into an engrossing tapestry of mystery and drama.

The complex realities of human emotions and experience are more than sufficient to carry a film, and Farhadi doesn't let fussy direction or attempts to be overly clever get in the way of letting this brilliant, quietly insightful story unfold.

A clarion cry that cuts through the xenophobic clutter and the dense fog of war to show that not everyone "over there" is a boogeyman waiting to jump out of the closet. If that sounds terribly simplistic, just consider where we live.

A Separation is more than a character study; it's a deftly plotted drama that unfolds layer after layer and will leave you asking all kinds of questions about your beliefs and moral judgments. Fascinating stuff.

The story has no winners, and the daringly ambiguous ending defies simple conclusions. Even the title can be taken multiple ways.

The movie opens the door on a foreign culture, yet the people do not seem that dissimilar. .... They deal with some of the same struggles ... -- generational and class conflicts, ideological differences, and connecting and communicating emotionally.

A devastating portrait of two families in crisis, with their class differences representing the title's true 'separation.'

"A Separation" deals with some very murky issues, but it does so with astonishing clarity.

The movie is a singular mix of the foreign and the familiar, but is it the best 2011 film in the whole wide world? A 99? I can't say that I concur with my colleagues on that score.

A Separation explores how presumed social norms are extremely tenuous and how threats to these almost illusory ideas can threaten our sense of personal security.
